Drug intelligence / Profile preview
rNa-GST-1 is a recombinant protein subunit vaccine candidate developed for the prevention of hookworm infection caused by *Necator americanus* and its associated complications, such as iron-deficiency anemia. The vaccine consists of a 24 kDa recombinant glutathione S-transferase-1 (Na-GST-1) antigen cloned from *N. americanus* and expressed in the yeast *Pichia pastoris*. Na-GST-1 is an essential enzyme in the hookworm's blood-feeding and hemoglobin digestion pathway, where it functions in heme and hematin detoxification. Immunization with rNa-GST-1, typically formulated with Alhydrogel (aluminum hydroxide) and adjuvants like CpG 10104 or GLA-AF, induces neutralizing antibodies that block the enzyme's catalytic and heme-binding activities, leading to parasite death or reduced fecundity. The vaccine has been evaluated in Phase 1 and Phase 2 clinical trials.
