RNA lipid particle aggregates (RNA-LPAs) are a novel cancer immunotherapy delivery system designed to enhance the payload packaging and immunogenicity of tumor mRNA antigens. Unlike conventional mRNA vaccine designs that package payload into nanoparticle cores for Toll-like receptor engagement in immune cells, RNA-LPAs feature an "onion-like" multi-lamellar structure that activates RIG-I in stromal cells when administered systemically. This activation elicits a massive cytokine/chemokine response and dendritic cell/lymphocyte trafficking that provokes cancer immunogenicity and mediates tumor rejection. The technology can be highly personalized to individual patients by extracting RNA from their tumors, amplifying it, and wrapping it into biocompatible lipid nanoparticles. RNA-LPAs have demonstrated the ability to rapidly transform "cold" tumors (with few immune cells and silenced immune response) into "hot" tumors (with active immune response) within 48 hours of administration.
