RNDO-Program 2 is an investigational immunotherapy program being developed by Rondo Therapeutics for the treatment of ovarian cancer. The program utilizes a dual-targeting strategy consisting of two distinct bispecific antibodies: one targeting CD3 and a tumor-associated antigen (TAA-1), and another targeting the costimulatory receptor CD28 and a second tumor-associated antigen (TAA-2). This approach is designed to provide both the primary activation signal (Signal 1 via CD3) and the costimulatory signal (Signal 2 via CD28) specifically within the tumor microenvironment. By requiring the presence of both tumor antigens for full T-cell activation, the therapy aims to selectively recruit and activate T cells against tumor cells in the immunosuppressive solid tumor microenvironment while potentially minimizing systemic toxicity associated with non-targeted costimulation. The program is currently in the IND-enabling stage of development.
