Overview

rNDV-TRAIL is a genetically engineered oncolytic virus therapy based on the Newcastle disease virus (NDV) backbone that has been modified to express the tumor necrosis factor-related apoptosis-inducing ligand (TRAIL). This recombinant agent combines the inherent tumor-selective and oncolytic properties of NDV with the pro-apoptotic activity of TRAIL. The mechanism involves infection of cancer cells by rNDV, which increases expression of death receptors (DR4 and DR5) and delivers TRAIL directly to tumor cells. TRAIL then binds these death receptors, triggering both intrinsic and extrinsic apoptotic pathways in malignant cells—including those resistant to conventional TRAIL therapy—resulting in enhanced cancer cell apoptosis. Preclinical studies have demonstrated superior efficacy for rNDV-TRAIL over parental NDV or TRAIL alone in colorectal cancer models, including those with resistance to standard TRAIL-induced apoptosis[1][6][7][10].
