Overview

Flindr Therapeutics is developing a first-in-class small molecule inhibitor of RNF31 (also known as HOIP), a protein-stabilizing E3 ubiquitin ligase that is a critical component of the Linear Ubiquitin Chain Assembly Complex (LUBAC). RNF31 is aberrantly activated in various solid and hematological malignancies, where it promotes cell survival. The inhibitor works by a dual mechanism: blocking the activation of the NF-κB signaling pathway and simultaneously inducing RIPK1-mediated programmed cell death. Currently in early clinical development, the program targets B-cell lymphomas and ovarian cancer, with a focus on using specific biomarkers to identify patients most likely to benefit from the therapy.
