rnf43-721 is a synthetic peptide derived from the ring finger protein 43 (RNF43), an oncoantigen highly expressed in various cancer cells, including colorectal cancer. It is used as one of two peptide epitopes in the investigational cancer vaccine OCV-C02, which also contains a TOMM34-derived peptide. The vaccine aims to induce cytotoxic T lymphocyte (CTL) responses against tumor cells expressing RNF43 and has been evaluated primarily for refractory metastatic colorectal cancer. Mechanistically, rnf43-721 acts as a tumor-associated antigen to stimulate immune responses when presented by HLA-A24-positive dendritic cells, leading to targeted anti-tumor immunity[3][2]. Clinical studies have shown that vaccination with this peptide can induce specific CTL responses and may contribute to disease stabilization in some patients[2][3].
