The RNF43 peptide vaccine is a therapeutic cancer vaccine composed of synthetic peptides derived from the ring finger protein 43 (RNF43), often combined with other tumor-associated antigen peptides such as TOMM34, VEGFR1, and VEGFR2. These vaccines are designed to stimulate cytotoxic T lymphocyte (CTL) responses against tumor cells expressing these antigens. The mechanism involves presenting HLA-A*2402-restricted epitopes to the immune system to induce a targeted anti-tumor immune response. The primary indication has been colorectal cancer, particularly in patients with refractory or metastatic disease and those who are HLA-A*2402-positive. Clinical trials have evaluated its safety and immunogenicity both as monotherapy and in combination with chemotherapeutic agents like tegafur/uracil plus leucovorin (UFT/LV).
