Overview

RNF5 inhibitors are a class of therapeutic agents designed to target and inhibit the E3 ubiquitin ligase RNF5 (also known as RMA1). RNF5 is a key regulator of the endoplasmic reticulum-associated degradation (ERAD) pathway, where it mediates the ubiquitination and subsequent degradation of various protein substrates. In oncology, particularly in acute myeloid leukemia (AML), RNF5 has been identified as a critical driver of leukemogenesis; its expression is often upregulated in AML patients and correlates with poor prognosis and treatment resistance. Preclinical studies have demonstrated that inhibition of RNF5 impairs the growth and survival of AML cells and significantly sensitizes them to histone deacetylase (HDAC) inhibitors. Beyond cancer, RNF5 inhibition is of interest in cystic fibrosis, as it regulates the degradation of the CFTR protein, and in various inflammatory and muscle-related disorders.
