Overview

RNJ1 is a tau-specific monoclonal antibody that binds to the amino-terminal epitope (amino acids 1–22) of both human and mouse tau proteins. It demonstrates enhanced reactivity to aggregated tau compared to other tau antibodies, such as HJ8.5 and RNF5. RNJ1 effectively neutralizes proteopathic tau seeds in vitro and has shown efficacy in reducing tau aggregation in biosensor cellular assays. In vivo, RNJ1 treatment improves motor function in the K3 mouse model of tauopathy, which expresses a mutant human tau associated with frontotemporal dementia. Additionally, both full IgG and scFv formats of RNJ1 can be delivered via mRNA to result in endogenous expression of functional antibody in human neuronal cells, supporting its potential as a therapeutic candidate for targeting tau pathology in neurodegenerative diseases such as Alzheimer’s disease and tauopathies[1][2].
