RNO-6 is a novel nitric oxide (NO)-donating ripasudil derivative designed to leverage the synergistic therapeutic effects of NO and Rho-associated protein kinase (ROCK) inhibitors for glaucoma treatment. It demonstrates potent ROCK inhibitory and NO-releasing capabilities, leading to F-actin depolymerization and suppressed mitochondrial respiration in human trabecular meshwork (HTM) cells. Preclinical studies in chronic ocular hypertension glaucoma mice showed that topical administration of RNO-6 significantly lowered intraocular pressure (IOP) and provided protection for visual function and retinal ganglion cells (RGCs), outperforming an equimolar dose of ripasudil. RNO-6 is being investigated as a promising agent for glaucoma or ocular hypertension.
