Overview

RNS60 is an investigational, physically-modified saline solution saturated with oxygen nanobubbles, developed by Revalesio. It is created using a proprietary process (Taylor-Couette-Poiseuille flow under elevated oxygen pressure) that generates charged oxygenated nanobubbles in 0.9% saline[1][6][7]. RNS60 is being developed as a potential treatment for neurological trauma and chronic neurodegenerative diseases driven by mitochondrial dysfunction, such as amyotrophic lateral sclerosis (ALS), acute ischemic stroke, traumatic brain injury (TBI), Alzheimer’s disease, and Parkinson’s disease[1][5][6][7]. The drug can be administered intravenously or via inhalation/nebulization[1][3]. Mechanistically, RNS60 activates intracellular signaling pathways—most notably the Type 1A PI3K pathway—which leads to anti-inflammatory effects and promotes mitochondrial biogenesis, cell survival, and differentiation[1][6][7]. It has demonstrated the ability to protect neurons and oligodendrocytes in the central nervous system while modulating immune cell activity to restore cellular homeostasis without typical side effects seen with many current medicines[1]. Preclinical studies have shown neuroprotective properties in models of ALS (including TDP-43 pathology), TBI, Alzheimer’s disease, and Parkinson’s disease; clinical trials have demonstrated safety and promising efficacy signals in ALS (slowed decline of respiratory function) and acute ischemic stroke (reduced infarct growth; improved functional outcomes)[2][4][5][8].
