RO-104 is a first-in-class, fully human tri-specific biologic antibody developed by RevOpsis Therapeutics for the treatment of retinal vascular diseases, particularly neovascular (wet) age-related macular degeneration (nAMD). It is engineered to simultaneously inhibit three clinically validated angiogenic pathways—vascular endothelial growth factor A (VEGF-A), vascular endothelial growth factor C (VEGF-C), and angiopoietin-2 (Ang-2)—which are implicated in the progression of retinal vascular disorders. By targeting all three pathways with a single molecule, RO-104 aims to improve visual outcomes and extend disease remission compared to current therapies that target only one or two pathways. The drug leverages RevOpsis' proprietary modular "plug-and-play" Rev-Mod platform for multispecific biologics discovery and development. As of June 2025, RO-104 remains in preclinical development with plans for first-in-human clinical trials in the United States[1][3][4][5][6].
