Ro 67-7476 is a potent and selective small molecule agonist of the metabotropic glutamate receptor 2 (mGlu2). Developed by Roche, it is primarily utilized as a pharmacological research tool to investigate glutamatergic neurotransmission and its role in central nervous system disorders such as schizophrenia, anxiety, and drug addiction. Activation of the mGlu2 receptor by Ro 67-7476 can trigger downstream signaling pathways, notably the extracellular signal-regulated kinase (ERK) pathway. In the context of oncology research, specifically in non-small-cell lung cancer (NSCLC) models, Ro 67-7476 has been used to induce ERK phosphorylation (p-ERK), effectively acting as a functional agonist of the ERK/MCL-1 pathway to study mechanisms of drug resistance and cell survival against chemotherapeutic agents like paclitaxel.
