Overview

Ro-CHOEP-21 is a combination chemotherapy regimen consisting of six agents: romidepsin (a histone deacetylase inhibitor), cyclophosphamide (an alkylating agent), doxorubicin (an anthracycline antibiotic), vincristine (a vinca alkaloid), etoposide (a topoisomerase II inhibitor), and prednisolone (a corticosteroid). This regimen was investigated as an induction therapy for untreated peripheral T-cell lymphomas (PTCLs) before high-dose chemotherapy and autologous stem cell transplantation. The addition of romidepsin to the standard CHOEP protocol aimed to improve progression-free survival by targeting both epigenetic regulation and multiple cellular pathways involved in lymphoma proliferation. However, clinical trials did not demonstrate improved progression-free survival compared to standard regimens[2][5][7].
