Overview

RO0730699 is a small molecule dipeptidyl peptidase-IV (DPP-IV) inhibitor developed by Hoffmann-La Roche for the treatment of type 2 diabetes mellitus. It functions by inhibiting the DPP-IV enzyme, which is responsible for the rapid degradation of the incretin hormones glucagon-like peptide-1 (GLP-1) and glucose-dependent insulinotropic polypeptide (GIP). By extending the half-life of these hormones, RO0730699 enhances glucose-dependent insulin secretion and reduces glucagon levels, thereby improving glycemic control. The compound was evaluated in Phase 2 clinical trials, including a randomized, double-blind, placebo-controlled study (NCT00114153) to assess its efficacy and safety in patients with type 2 diabetes. However, the drug did not progress to Phase 3 trials or regulatory approval and is no longer active in Roche's development pipeline.
