RO3300074 is an orally bioavailable small molecule inhibitor of matrix metalloproteinases 9 (MMP-9) and 12 (MMP-12), developed by Roche for the treatment of chronic obstructive pulmonary disease (COPD) and pulmonary emphysema. Matrix metalloproteinases are proteolytic enzymes that degrade extracellular matrix components; MMP-9 and MMP-12 are specifically associated with the inflammatory response and lung tissue destruction seen in smoking-related emphysema. The compound was investigated in a Phase 2, double-blind, placebo-controlled trial (NCT00431210) to evaluate its efficacy and safety in ex-smokers with moderate-to-severe COPD over a two-year period. Despite completing Phase 2, the program does not appear to have progressed to Phase 3 and is considered discontinued.
