RO5200628 is a small molecule triple reuptake inhibitor (SNDRI) that was developed by Hoffmann-La Roche for the treatment of major depressive disorder (MDD). It is designed to simultaneously inhibit the reuptake of three key monoamine neurotransmitters—serotonin, norepinephrine, and dopamine—from the synaptic cleft into the presynaptic neuron. By increasing the synaptic availability of all three neurotransmitters, RO5200628 was intended to provide enhanced therapeutic efficacy and a more rapid onset of antidepressant action compared to selective serotonin reuptake inhibitors (SSRIs) or dual reuptake inhibitors. Clinical development included Phase 1 safety and pharmacokinetic studies, such as an excretion balance study in healthy volunteers, as well as Phase 2 trials in patients with MDD. However, development of the compound was discontinued by Roche and it is no longer listed in their active pipeline.
