Drug intelligence / Profile preview
RO5256390 is a preclinical small molecule agonist of the trace amine-associated receptor 1 (TAAR1) developed by Hoffmann-La Roche. It acts as a full agonist at human, rat, and monkey TAAR1, and as a partial agonist at mouse TAAR1. By activating TAAR1, RO5256390 modulates monoaminergic neurotransmission, including dopamine, serotonin, and norepinephrine signaling. In preclinical models, it has demonstrated antipsychotic-like, pro-cognitive, and antidepressant-like effects, as well as efficacy in reducing cocaine reinforcement, alcohol consumption, and compulsive binge-eating behavior. Additionally, RO5256390 has been shown to bind directly to the dopamine transporter (DAT) and inhibit dopamine uptake.
