Overview

RO5468924 is the major pharmacologically active metabolite of alectinib, a small molecule tyrosine kinase inhibitor. It is generated in vivo after administration of alectinib and retains similar in vitro biological activity against its target, namely the rearranged during transfection (RET) tyrosine kinase, and is responsible for a substantial share of alectinib’s clinical efficacy. The metabolite is characterized by similar pharmacokinetic and pharmacodynamic properties to the parent compound and is monitored in plasma alongside alectinib in pharmacokinetic studies[1][3].
