RO6870868 is an oral prodrug developed by Hoffmann-La Roche that is rapidly converted in the body to RO6871765, a specific agonist of toll-like receptor 7 (TLR7). TLR7 agonists stimulate the innate immune system by activating interferon pathways and other immune responses. The drug was investigated primarily for its potential to treat chronic hepatitis B infection, aiming to enhance host antiviral immunity. In phase 1 clinical trials involving healthy volunteers, single oral doses of RO6870868 were generally well-tolerated and showed dose-dependent pharmacodynamic activity consistent with TLR7 engagement. However, development for infectious diseases was discontinued after phase 1.
