Overview

RO6874813 (also known as RG7386) is a bispecific antibody developed by Hoffmann-La Roche for the treatment of locally advanced or metastatic solid tumors. It is designed to simultaneously bind fibroblast activation protein-alpha (FAPα), which is highly expressed on cancer-associated fibroblasts in the tumor microenvironment, and death receptor 5 (DR5), a member of the TNF superfamily found on tumor cells. By binding both targets, RO6874813 induces apoptosis selectively in tumor cells through DR5 activation while leveraging FAP-mediated localization within the tumor stroma. The drug was administered intravenously and reached Phase 1 clinical trials for sarcoma and other solid tumors but its development has been discontinued[1][2][5][6][8].
