Overview
RO7020322 (also known as RG7834) is a highly selective and orally bioavailable small molecule inhibitor developed by F. Hoffmann-La Roche for the treatment of chronic hepatitis B virus (HBV) infection. It acts as an inhibitor of PAPD5 and PAPD7, which are non-canonical RNA polymerases involved in HBV mRNA processing. By inhibiting these targets, RO7020322 potently suppresses the production of HBV antigens (including HBsAg and HBeAg) and viral DNA. The drug demonstrated nanomolar potency in preclinical models but was discontinued after reaching Phase 1 clinical trials[1][3][6][8].
