RO7114667 is a monoclonal antibody developed by Hoffmann La Roche that specifically neutralizes interleukin-1 beta (IL-1β), a key proinflammatory cytokine involved in the sterile inflammation that drives the progression of chronic kidney disease (CKD), particularly in the context of type 2 diabetes. The antibody was engineered as a monoclonal antibody in a mouse hybridoma and reformatted with a murine IgG1 scaffold featuring effector-silencing DAPG mutations. It demonstrates strong cross-reactivity between human and murine IL-1β but does not bind to IL-1α. Preclinical studies in uninephrectomized diabetic db/db mice have shown that RO7114667 can preserve renal function by significantly attenuating the decline of the glomerular filtration rate (GFR) and protecting podocyte numbers, without altering systemic metabolic parameters such as blood glucose or body weight.
