RO7172508 (also known as RG6123) is a bispecific T-cell engaging monoclonal antibody developed by Roche. It targets carcinoembryonic antigen-related cell adhesion molecule 5 (CEA, also known as CEACAM5) expressed on tumor cells and the cluster of differentiation 3 epsilon chain (CD3 epsilon chain) on T-cells. By binding both CEA and CD3, it redirects T-cells to attack CEA-expressing tumor cells. Compared to earlier agents like cibisatamab (RO6958688), RO7172508 was designed for higher affinity to CEA, increased potency, and a longer half-life. The drug was investigated in patients with locally advanced or metastatic solid tumors that express CEA but development was discontinued after phase I trials due to lack of efficacy[1][3][4].
