RO7284755 (also known as RG6325) is an investigational recombinant fusion protein developed by Genentech (Roche) for the treatment of hematologic malignancies, specifically relapsed or refractory multiple myeloma. The molecule consists of human interleukin-15 (IL-15) non-covalently bound to the sushi domain of the IL-15 receptor alpha (IL-15Rα) chain, which is fused to a human IgG1 Fc domain. This configuration acts as an IL-15 superagonist, mimicking the natural trans-presentation of IL-15 by dendritic cells to effector lymphocytes. By complexing IL-15 with its alpha receptor, the drug exhibits significantly enhanced biological activity and a prolonged circulatory half-life compared to monomeric IL-15. RO7284755 selectively stimulates the proliferation and activation of natural killer (NK) cells and CD8+ cytotoxic T cells, which are critical for anti-tumor immunity, while minimizing the activation of regulatory T cells (Tregs) that can occur with IL-2 therapy. It is currently being evaluated in Phase 1 clinical trials as both a monotherapy and in combination with other agents like daratumumab.
