RO7297089 is a bispecific tetravalent antibody that targets both B-cell maturation antigen (BCMA) and CD16a (FcγRIIIa). BCMA is highly expressed on plasmablasts and plasma cells and is overexpressed in malignant plasma cells associated with multiple myeloma. CD16a is a transmembrane receptor found on natural killer (NK) cells, mast cells, macrophages, and monocyte subsets. By binding to BCMA on myeloma cells and CD16a on innate immune effector cells such as NK cells, RO7297089 acts as an innate cell engager designed to induce lysis of BCMA-positive multiple myeloma tumor cells through antibody-dependent cellular cytotoxicity. The drug was developed using Affimed’s ROCK platform technology by Affimed Therapeutics and further developed by Genentech/Roche for the treatment of relapsed or refractory multiple myeloma[1][2][3][4][5][7][8].
