It targets both fibroblast activation protein alpha (FAP), which is highly expressed in the tumor microenvironment of most solid tumors, and CD40, a receptor found on antigen-presenting immune cells. By binding to FAP on tumor stroma and CD40 on immune cells simultaneously, RO7300490 localizes its agonist activity to the tumor site. This dual targeting aims to activate immune responses against cancer cells while minimizing systemic toxicities associated with conventional CD40 agonists. The drug was developed for use as a single agent or in combination with atezolizumab (an anti-PD-L1 antibody) for advanced or metastatic solid tumors[1][2][3][6][7].
