RO7303359 is an investigational anti-interleukin 33 (IL-33) fragment antigen-binding region (Fab) monoclonal antibody developed for the treatment of geographic atrophy secondary to age-related macular degeneration (AMD). It was designed to target the IL-33/ST2 pathway, a proinflammatory signaling axis implicated in AMD pathophysiology. The drug was administered as a single intravitreal injection in Phase 1 clinical trials to evaluate its safety, tolerability, pharmacokinetics, and immunogenicity. While it demonstrated an acceptable safety profile and dose-proportional pharmacokinetics up to 20 mg per eye, no significant effect on IL-33/ST2 pathway biomarkers was observed in aqueous humor samples. As a result of these findings and lack of demonstrable pharmacodynamic activity on its intended target pathway in humans with geographic atrophy due to AMD, further development for this indication has been discontinued[8][4][2].
