Overview

RO7308480 (also known as NTX-2001) is an oral small molecule agonist of the trace amine-associated receptor 1 (TAAR1). Originally developed by Roche and subsequently licensed to Newleos Therapeutics, the compound is being investigated for the treatment of neuropsychiatric conditions, specifically substance use disorders. TAAR1 is a G protein-coupled receptor that modulates dopaminergic, serotonergic, and glutamatergic neurotransmission in the central nervous system. By activating TAAR1, RO7308480 aims to regulate these pathways without the side effects typically associated with direct receptor antagonists or stimulants. It has undergone Phase 1 clinical evaluation for safety, pharmacokinetics, and drug-drug interactions, including studies on its interaction with midazolam.
