Overview

RO7443904 is a bispecific antibody designed to target both CD19 on B cells and CD28 on T cells. It acts as a costimulatory agonist by binding to these two antigens, delivering a strong costimulatory signal (signal 2) that augments and prolongs T-cell activity after initial activation by a tumor-targeting T-cell engaging bispecific antibody (signal 1). This mechanism is intended to enhance the immune response against B-cell malignancies such as non-Hodgkin lymphoma. RO7443904 has been developed primarily for use in combination with glofitamab, another bispecific antibody targeting CD20 and CD3. The combination aims to provide an "off-the-shelf" alternative to CAR-T therapy with potentially less toxicity than first-generation CD28 agonists. Clinical trials have shown that this approach can boost T-cell proliferation and effector function in patients with relapsed or refractory non-Hodgkin lymphoma[1][5][6].
