RO7444973 is a bispecific T-cell engaging antibody developed by Roche (Hoffmann-La Roche) that targets both the tumor-associated antigen human leukocyte antigen (HLA)-A2-restricted, melanoma-associated antigen A4 (MAGE-A4), and the T-cell surface antigen CD3. Its mechanism of action involves redirecting T cells to recognize and kill MAGE-A4-positive tumor cells in patients carrying the HLA-A*02:01 allele. The drug was investigated for use in unresectable and/or metastatic solid tumors expressing MAGE-A4, but its clinical development was terminated after a phase 1 trial[1][2][6].
