Overview

RO7446603 + aflibercept is an investigational **combination therapy** currently in clinical trials for the treatment of diabetic macular edema. RO7446603 is being developed as a Tie-2 agonist, which targets the angiopoietin/Tie-2 pathway, playing a role in vascular stabilization and leakage. Aflibercept is a well-established VEGF (vascular endothelial growth factor) inhibitor that binds and neutralizes VEGF-A, VEGF-B, and placental growth factor, reducing abnormal blood vessel proliferation and permeability. The combination is administered as separate intravitreal injections in early-stage trials, with the aim of improving vascular stability and visual outcomes by simultaneously modulating both Tie-2 and VEGF pathways. Genentech (a Roche subsidiary) is leading the development of the combination in phase 1/2 clinical studies for diabetic macular edema. The combination has not been approved for any indication.
