Overview

RO7494222 (also known as RG6356) is an investigational humanized monoclonal antibody developed by Roche for the treatment of Duchenne muscular dystrophy (DMD). It functions as a myostatin inhibitor, specifically targeting and neutralizing myostatin (Growth Differentiation Factor 8), a negative regulator of muscle growth. By inhibiting myostatin signaling, RO7494222 was intended to promote muscle hypertrophy and prevent muscle wasting, potentially improving physical function and slowing disease progression in patients with DMD. The drug reached Phase 1 clinical evaluation, where its safety, tolerability, and pharmacokinetics were assessed in both healthy participants and patients with DMD. However, Roche discontinued the development of RO7494222 in 2023 and removed it from its active pipeline.
