Overview

RO7496353 + nivolumab + oxaliplatin + capecitabine is an investigational combination regimen for advanced or metastatic solid tumors. RO7496353 is a monoclonal antibody that selectively inhibits the activation of latent transforming growth factor-beta 1 (TGF-β1), aiming to counteract the immunosuppressive tumor microenvironment and improve anti-tumor immunity. Nivolumab is a monoclonal antibody that inhibits programmed death-1 (PD-1), enhancing T-cell response against cancer cells. Oxaliplatin is a platinum-based chemotherapeutic agent causing DNA cross-linking and apoptosis in rapidly dividing cells. Capecitabine is an orally administered prodrug of 5-fluorouracil (5-FU), inhibiting thymidylate synthase and DNA synthesis in tumor cells. The combination is being evaluated for synergistic efficacy in solid tumors, especially in settings such as non-small cell lung cancer, gastric cancer, and pancreatic ductal adenocarcinoma, as part of an early-phase clinical development program led by Genentech/Roche. The regimen is typically administered via intravenous infusion (IV) for RO7496353, nivolumab, and oxaliplatin, and orally for capecitabine[1][2][3][5].
