Overview

RO7504109 (also known as RG6526) is an investigational humanized monoclonal antibody fragment (Fab) developed by Roche for the treatment of geographic atrophy (GA) secondary to age-related macular degeneration (AMD). It is designed to inhibit complement factor B (CFB), a key serine protease in the alternative complement pathway. By targeting Factor B, RO7504109 prevents the formation of the C3 convertase (C3bBb), thereby inhibiting the amplification loop of the complement cascade and reducing the inflammatory damage to the retinal pigment epithelium and photoreceptors. The drug is administered via intravitreal injection and is currently being evaluated in Phase 2 clinical trials (e.g., the GALILEO study) to assess its efficacy in slowing the progression of GA lesions.
