Overview

RO7567132 is a bispecific antibody developed by Roche (Hoffmann-La Roche) for the treatment of advanced and metastatic solid tumors. It targets both fibroblast activation protein alpha (FAP), which is expressed on cancer-associated fibroblasts in the tumor stroma, and lymphotoxin beta receptor (LTBR; also known as TNFRSF3), which is present on various cells in the tumor microenvironment and vasculature. The drug acts as an antagonist to FAP and an agonist to LTBR/TNFRSF3. Preclinical studies have shown that RO7567132 can convert immunologically “cold” tumors into “hot” ones, thereby enhancing responses to immune checkpoint inhibitors and T-cell engager therapies. It is currently being evaluated in phase 1 clinical trials both as a single agent and in combination with atezolizumab for patients with advanced or metastatic solid cancers who have exhausted standard treatment options[3][4][9].
