Overview

RO7624781 (also known as RG6538) is an investigational recycling monoclonal antibody designed to target C1s, a serine protease within the C1 complex of the classical complement pathway. Developed by Genentech (a member of the Roche Group) using pH-dependent "recycling" antibody technology, RO7624781 binds to C1s in the systemic circulation and releases it in the acidic environment of the endosome for lysosomal degradation. The antibody then recycles back to the cell surface via the neonatal Fc receptor (FcRn), allowing a single antibody molecule to neutralize multiple C1s molecules. This mechanism is intended to provide more potent and durable inhibition of the classical complement pathway compared to conventional antibodies. RO7624781 is currently in Phase 1 clinical development for the treatment of complement-mediated immunological disorders, including cold agglutinin disease (CAD).
