RO7694744 is a trivalent bispecific antibody-cytokine fusion protein developed by Roche using the Dutaflip platform. It is designed to target Interferon alpha-2a (IFN-α2a) specifically to cells expressing Programmed cell death ligand 1 (PD-L1). This targeted approach aims to concentrate the immunomodulatory effects of IFN-α within the tumor microenvironment (TME), thereby enhancing anti-tumor immune responses and inducing inflammatory gene signatures in PD-L1-positive tumor and immune cells while minimizing the systemic toxicity typically associated with non-targeted interferon therapy. Preclinical evaluations using patient-derived tumor explants (PTEs) from non-small cell lung cancer (NSCLC) and bladder cancer (BLCA) have demonstrated that RO7694744 induces a consistent and specific cytokine response (including IP-10, ISG-15, and IFN-γ) and preferentially activates PD-L1-positive cellular subpopulations.
