RO7790118 (also known as RG6511) is a bispecific T-cell engager (TCE) monoclonal antibody developed by Roche for the treatment of advanced hepatocellular carcinoma (HCC). The molecule is engineered to simultaneously target Glypican-3 (GPC3), a cell-surface oncofetal antigen highly expressed in HCC, and the CD3 epsilon subunit of the T-cell receptor complex on T-lymphocytes. By facilitating the formation of an immunological synapse between tumor cells and cytotoxic T-cells, RO7790118 induces T-cell activation, cytokine release, and directed lysis of GPC3-positive cancer cells. This approach aims to leverage the patient's immune system to specifically target and eliminate liver cancer cells while minimizing off-target effects. RO7790118 is currently being evaluated in Phase 1 clinical trials to determine its safety, tolerability, pharmacokinetics, and preliminary anti-tumor activity in patients with GPC3-expressing tumors.
