Overview

RO7795081 + itraconazole refers to the co-administration of the investigational compound RO7795081 (also known as RG6356) and the antifungal agent itraconazole in clinical pharmacology studies. RO7795081 is a small molecule antagonist of the G protein-coupled receptor 55 (GPR55), developed by Roche as a potential dystrophin-independent therapy for Duchenne muscular dystrophy (DMD). The combination was specifically utilized in Phase 1 drug-drug interaction (DDI) trials to evaluate the effect of itraconazole, a potent inhibitor of the cytochrome P450 3A4 (CYP3A4) enzyme, on the pharmacokinetics and safety profile of RO7795081. In late 2022, Roche announced the discontinuation of the RO7795081 (RG6356) program following a review of clinical data.
