Drug intelligence / Profile preview
RO7821657 is an investigational protease-activated T-cell bispecific (TCB) antibody developed by Roche for the treatment of locally advanced and/or metastatic solid tumors, specifically microsatellite-stable (MSS) colorectal cancer (CRC) and non-squamous non-small cell lung cancer (NSCLC). The antibody is designed to target Carcinoembryonic Antigen-Related Cell Adhesion Molecule 5 (CEACAM5) on tumor cells and CD3 on T cells. By incorporating a protease-cleavable linker/mask, the T-cell engaging activity of RO7821657 is conditionally restricted to the tumor microenvironment where tumor-associated proteases are active, thereby minimizing systemic toxicity and sparing healthy tissues that express CEACAM5.
