Overview

RO7823653 is an investigational monoclonal antibody developed by Genentech (a member of the Roche Group) for the treatment of diabetic macular edema (DME). It specifically targets and neutralizes interleukin-6 (IL-6), a pro-inflammatory cytokine that is significantly involved in the pathogenesis of retinal vascular diseases, including increased vascular permeability and chronic inflammation. By inhibiting IL-6 signaling, RO7823653 aims to reduce macular edema and improve visual outcomes in patients who may not respond optimally to standard anti-VEGF therapies. The drug is currently being evaluated in a Phase I clinical trial (NCT06121401) to assess its safety, pharmacokinetics, and pharmacodynamics as both a monotherapy and in combination with faricimab (Vabysmo), a bispecific antibody targeting VEGF and Ang-2.
