Roasted licorice extract (rLE) is a processed botanical extract derived from the dried roots of *Glycyrrhiza* species. The roasting process significantly alters the chemical profile of the extract, notably increasing the concentration of licochalcone A (up to five-fold) while reducing levels of glycyrrhizin and isoliquiritigenin. Preclinical studies indicate that rLE possesses potent anti-inflammatory and anti-arthritic properties, demonstrating superior efficacy compared to unroasted licorice in acute inflammation models. Its therapeutic effects are mediated through the suppression of pro-inflammatory cytokines, including tumor necrosis factor-alpha (TNF-α) and interleukin-1beta (IL-1β), and the inhibition of matrix metalloproteinase-3 (MMP-3) expression in joint tissues. Additionally, rLE exhibits antioxidant activity by reducing lipid peroxidation and restoring endogenous antioxidant defenses such as glutathione and catalase, thereby protecting against oxidative tissue damage in chronic inflammatory conditions like rheumatoid arthritis.
