Robalzotan is a small molecule investigational drug developed as a selective antagonist of the serotonin 5-hydroxytryptamine 1A (5‑HT₁A) receptor. It binds with high affinity and selectivity to the 5‑HT₁A receptor, blocking its activity. This mechanism was intended to enhance serotonergic neurotransmission by preventing autoreceptor-mediated inhibition of serotonin release, potentially augmenting the effects of selective serotonin reuptake inhibitors (SSRIs) or serving as monotherapy in depression and anxiety disorders. Robalzotan was also studied for gastrointestinal indications such as irritable bowel syndrome (IBS). Despite promising preclinical pharmacology and CNS penetration, clinical trials did not demonstrate sufficient efficacy or safety; development was discontinued after phase II studies due to lack of benefit and adverse central nervous system effects[2][6][8].
