Robatumumab + everolimus is an investigational combination therapy composed of robatumumab, a fully human monoclonal antibody targeting insulin-like growth factor receptor-1 (IGF-1R), and everolimus, a small molecule inhibitor of mammalian target of rapamycin (mTOR). Robatumumab inhibits IGF-1R signaling, impeding cancer cell growth and survival, while everolimus blocks mTOR, a key regulator of cell proliferation and angiogenesis. The combination has been studied preclinically and clinically for synergistic antitumor activity, particularly in advanced sarcomas and renal cell carcinoma. Robatumumab was developed by Schering-Plough (later Merck), and everolimus by Novartis. The combination has been evaluated in at least phase I/II trials, but development of robatumumab was terminated and the combination is not approved for any indication[3][1][5].
