Overview

Robatumumab + irinotecan is an investigational combination therapy evaluated by Merck Sharp & Dohme for the treatment of advanced solid tumors. Robatumumab (SCH 717454, MK-7454) is a fully human monoclonal antibody that targets the insulin-like growth factor-1 receptor (IGF-1R), inhibiting signaling pathways that drive tumor cell growth and survival. Irinotecan is a small molecule chemotherapy agent that inhibits DNA topoisomerase I, leading to double-strand DNA breaks and cell death. This combination was studied in a Phase 1B/2 trial (NCT00954512) in patients with colorectal adenocarcinoma, non-small cell lung cancer, gastric adenocarcinoma, breast cancer, renal cell carcinoma, and pancreatic adenocarcinoma. The trial was ultimately terminated, and development of the combination was discontinued.
