ROBO1 CAR-NK cells are a form of cell therapy in which natural killer (NK) cells are genetically engineered to express a chimeric antigen receptor (CAR) that specifically targets the Roundabout homolog 1 (ROBO1) protein. ROBO1 is an axon guidance receptor overexpressed in several solid tumors, including ovarian and pancreatic cancers. The therapy involves isolating NK cells—often from peripheral blood mononuclear cells (PBMCs)—and modifying them to recognize and attack tumor cells expressing ROBO1. Preclinical studies have demonstrated that these modified NK cells exhibit enhanced cytotoxicity against primary tumor samples and organoids derived from patients with ovarian cancer[1], as well as efficacy in pancreatic cancer models[2][5]. This approach is being developed both as autologous personalized immunotherapy and as allogeneic "off-the-shelf" products using established NK cell lines such as NK92[5]. The main mechanism of action is targeted lysis of tumor cells via recognition of the ROBO1 antigen on their surface.
