Overview

ROBO4 short hairpin RNA (shRNA) is an experimental RNA interference (RNAi) therapeutic candidate designed to silence the expression of Roundabout homolog 4 (ROBO4). While ROBO4 is traditionally recognized for its role in endothelial cells and angiogenesis, recent research has identified its expression in epithelial breast cancer cells, particularly in basal-like tumors. By targeting ROBO4, this shRNA reduces the population of hybrid epithelial-mesenchymal stem-like cells, thereby inhibiting tumor growth, colony formation, and cellular plasticity. Preclinical studies using patient-derived xenograft (PDX) models have demonstrated that both constitutive and inducible knockdown of ROBO4 can significantly reduce tumorigenesis and halt the growth of established tumors. The cell-surface location of ROBO4 makes it an attractive target for potential therapeutic applications in breast cancer.
