Robotnikinin is a small molecule that functions as a potent modulator of the Sonic Hedgehog (SHH) signaling pathway. Originally identified through high-throughput screening at the Broad Institute, it is recognized for its unique ability to bind directly to the Sonic Hedgehog protein (ShhN), thereby preventing the ligand from interacting with its receptor, Patched. In the context of pancreatic ductal adenocarcinoma (PDAC), research indicates that robotnikinin acts as an agonist of Patched 2 (Ptch2), a tumor suppressor within the SHH pathway. By activating Ptch2, robotnikinin helps to counteract the pathway's overactivation, which is often driven by fibroblast-derived extracellular vesicles in the tumor microenvironment. This modulation has been shown to reduce tumor organoid growth and reverse chemoresistance to agents like gemcitabine in preclinical models of primary and metastatic pancreatic cancer.
