ROC0929 is a selective small molecule inhibitor of secreted phospholipase A2 group X (sPLA2-X), an enzyme encoded by the PLA2G10 gene that is implicated in the pathogenesis of asthma and allergic inflammation. Developed by researchers at the University of Washington, ROC0929 is designed to block the enzymatic activity of sPLA2-X, which acts as an endogenous adjuvant for type-2 immune responses and promotes peripheral sensitization to allergens such as house dust mite. In preclinical studies, ROC0929 has demonstrated the ability to attenuate airway hyperresponsiveness (AHR) and reduce the recruitment of allergen-specific CD4+ T-cells and eosinophils to the lungs when administered during the sensitization phase. This mechanism suggests a potential therapeutic role for ROC0929 in preventing the "atopic march" and treating chronic allergic airway diseases.
